All keys lost unable to get isn't because I have no ignition. Can someone help me thanks

What is DME?
If it is MSX8X then connect cas and dme on bench
After you read ISN from DME, you can return them back to car and make the key.

Thanks. The dme is msd81, cas3. Do I need to connect the key slot?

Is saying can not get vo.

I connected the key slot. Still can't read vo.

No need, because you will provide ignition ON to the DME via 12v supply to the ignition pin in the DME.
You can also provide key slot if available, but you must have a working key in this case, besides to many extra wiring between DME and CAS.

Cas pin 12 - ground, 26 - can, 35 - canH, 34 - 12v.
Dme 4 - ground, 1,1- 12v, 44 -canL, 28 - canH.

On my new bench cable main positive goes to pin 16. Device power is connected to pin1. But 16 on 1 are not interconnected, so I have no voltage to my modules.

You cannot use the Tricore boot mode cable to do this job.
Autohex II controls the powering to modules via Pin 1, this is available only when you work in ISN boot mode software.
You must make your own cable for this job.

I'm almost there. I made a new cable. I can communicate with the dme and cas. When I go to read the isn, the software is asking me to program the dme with the ignition off. This it ok to say yes? Thanks.

If you provide 12V to DME ignition pin, then just go forward :)
